Do I need to prime my autolube? It's been unused for the past 2 years (previous owner) and I want to start using it again, wasn't sure if I had to prime it or how to tell if it works at all?

If it's been disconnected or the pipe blocked then yes, pull the pipe off at the carb box and let the oil drip, pop the pipe back on and you will see an air bubble in the pipe. Run the scooter for one tank still with oil in the mix, if the autolube is working the bubble will disappear and you can on the next tank start using autolube.
For a while on that first tank you will be running too much oil, but that will work through.
Be sure the autolube is working though, prior owner may have disconnected for a reason other than to run a different mix, or because he didn't want to rely on autolube.

I was informed that it might be disconnected but the pipe is still on (not blocked) the worm drive is still in, and it still has autolube gasket so how could it be disconnected?

And the oil tank is empty?
I'd put some oil in the tank, prime like I suggest and you will know if it's working or not.. if the air bubble goes nowhere after a few days use its not working.
